Circuit formation and function in the olfactory bulb of mice with reduced spontaneous afferent activity.

چکیده
The type of neuronal activity required for circuit development is a matter of significant debate. We addressed this issue by analyzing the topographic organization of the olfactory bulb in transgenic mice engineered to have very little afferent spontaneous activity due to the overexpression of the inwardly rectifying potassium channel Kir2.1 in the olfactory sensory neurons (Kir2.1 mice). In these conditions, the topography of the olfactory bulb was unrefined. Odor-evoked responses were readily recorded in glomeruli with reduced spontaneous afferent activity, although the functional maps were coarser than in controls and contributed to altered olfactory discrimination behavior. In addition, overexpression of Kir2.1 in adults induced a regression of the already refined connectivity to an immature (i.e., coarser) status. Our data suggest that spontaneous activity plays a critical role not only in the development but also in the maintenance of the topography of the olfactory bulb and in sensory information processing.

P11: Assess the Electrophysiological Activity of Olfactory Bulb in the Animal Model of PTSD and its Relationship with Neuroinflammation in the Olfactory Bulb
Post-traumatic stress disorder (PTSD) is a mental health condition that's triggered by a terrifying event - either experiencing it or witnessing it. In addition to the relationship between PTSD and neuroinflammation, research indicates that olfactory bulb are effective in anxiety disorders. The aim of this study was to assess the electrophysiological activity of olfactory bulb in the animal mod...
متن کاملZinc-finger gene Fez in the olfactory sensory neurons regulates development of the olfactory bulb non-cell-autonomously.
Fez is a zinc-finger gene encoding a transcriptional repressor that is expressed in the olfactory epithelium, hypothalamus, ventrolateral pallium and prethalamus at mid-gestation. To reveal its function, we generated Fez-deficient mice. The Fez-deficient mice showed several abnormalities in the olfactory system: (1) impaired axonal projection of the olfactory sensory neurons; (2) reduced size o...

The olfactory system provides mammals with the abilities to investigate, communicate and interact with their environment. These functions are achieved through a finely organized circuit starting from the nasal cavity, passing through the olfactory bulb and ending in various cortical areas. We show that the absence of transient axonal glycoprotein-1 (Tag1)/contactin-2 (Cntn2) in mice results in ...
متن کاملThe Effects of Soy Milk on Histomorphometric Changes of Olfactory Bulb in Neonatal Ovariectomized RatsSprague- Dawley strain
Background & Objective: Soy milk contains isoflavones that comprises the phytoestrogens families. They have structural similarities with mammalian estrogen. This study was done to investigate the effects of soy milk on histomorphometric changes of olfactory bulb in neonatal ovariectomized rats. Materials & Methods: Thirty female rats Sprague- Dawley strain (one-day old) were kept in a standa...
متن کاملNrp2 is sufficient to instruct circuit formation of mitral-cells to mediate odour-induced attractive social responses
Odour information induces various innate responses that are critical to the survival of the individual and for the species. An axon guidance molecule, Neuropilin 2 (Nrp2), is known to mediate targeting of olfactory sensory neurons (primary neurons), to the posteroventral main olfactory bulb (PV MOB) in mice. Here we report that Nrp2-positive (Nrp2+) mitral cells (MCs, second-order neurons) play...
